Endothelial cell autophagy in homeostasis and cancer

چکیده

Autophagy, the major lysosomal pathway for degradation and recycling of cytoplasmic materials, is increasingly recognized as a player in endothelial cell (EC) biology vascular pathology. Particularly solid tumors, tumor microenvironmental stress such hypoxia, nutrient deprivation, inflammatory mediators, metabolic aberrations stimulates autophagy tumor-associated blood vessels. Increased ECs may serve mechanism to alleviate restrict exacerbated responses. However, increased can re-model pathways affect trafficking surface availability key mediators regulators interplay between EC immune cells. In line with this, heightened involved pathological angiogenesis, inflammatory, Here, we review cellular molecular mechanisms regulated by under physiological conditions discuss recent evidence implicating angiogenesis immunosurveillance.

ER homeostasis and autophagy

The endoplasmic reticulum (ER) is a key site for lipid biosynthesis and folding of nascent transmembrane and secretory proteins. These processes are maintained by careful homeostatic control of the environment within the ER lumen. Signalling sensors within the ER detect perturbations within the lumen (ER stress) and employ downstream signalling cascades that engage effector mechanisms to restor...

Autophagy, cell death, and cancer

Autophagy is an evolutionarily conserved intracellular catabolic process that is used by all cells to degrade dysfunctional or unnecessary cytoplasmic components through delivery to the lysosome. Increasing evidence reveals that autophagic dysfunction is associated with human diseases, such as cancer. Paradoxically, although autophagy is well recognized as a cell survival process that promotes ...

Mevalonate Pathway Regulates Cell Size Homeostasis and Proteostasis through Autophagy

Balance between cell growth and proliferation determines cell size homeostasis, but little is known about how metabolic pathways are involved in the maintenance of this balance. Here, we perform a screen with a library of clinically used drug molecules for their effects on cell size. We find that statins, inhibitors of the mevalonate pathway, reduce cell proliferation and increase cell size and...

Mechanotransduction and endothelial cell homeostasis: the wisdom of the cell.

Vascular endothelial cells (ECs) play significant roles in regulating circulatory functions. Mechanical stimuli, including the stretch and shear stress resulting from circulatory pressure and flow, modulate EC functions by activating mechanosensors, signaling pathways, and gene and protein expressions. Mechanical forces with a clear direction (e.g., the pulsatile shear stress and the uniaxial c...

Cell-mediated autophagy promotes cancer cell survival.

Immune effector cells integrate signals that define the nature and magnitude of the subsequent response. Experimental measures for immune cell-mediated lysis of tumors or virally infected targets rely on average responses of permeability or apoptotic changes within a population of targets. Here, we examined individual target cells following interaction with lymphoid effectors. We found that hum...
